: 단백질을 크기별로 분리하는 기술로 항원-항체 reaction(반응)을 이용하여 전체 단백질에서 특정 단백질만을 Detection 할 수 있다
이번 Western Blotting은 뉴런으로 분화하기 전인 Neuroblastoma(Ne,신경모세포)를 사용해 신경모세포가 암이 되게 유발하는 단백질을 찾을 수 있다
-test(실험) 방법
1) Sampling (Protein Extraction)
추출해 낼 세포에 Lysis buffer를 넣고 4도씨에서 원심 분리를 한다.50ml 튜브에 폴리아크릴아마이드 3.4ml, TDS와 SDS를 합하여 2.5ml, D.W 4.1ml를 넣고 혼합한 뒤 membrane 사이에 넣어준다.
Lane을 표시할 comb를 넣는다.
2) SDS-PAGE (sodium dodecyl sulfate - poly acrylamid gel elextrophoresis)

inner membrane과 outer membrane을 준비해 캐스터에 끼운 후 membrane 사이에 증류수를 넣어 물질이 새는지 시간을 두고 watch한 뒤 새지 않으면 증류수는 버린다.
Gel을 굳힐 때는 10% AP(Ammonium Persulfate와 TEMED를 사용한다.
